B I09 is a small-molecule inhibitor of the IRE-1 RNase with a reported in vitro IC50 of 1,230 nM. It inhibits splicing of XBP1 mRNA and reduces expression of XBP-1s in cellular assays, and is used to probe IRE-1-mediated unfolded protein response signaling. The compound is a light yellow to brown solid, soluble in DMSO, and unstable in solution so solutions should be freshly prepared.
