Baird Parker Agar with Egg Yolk Tellurite is a selective and differential medium used for the detection and enumeration of coagulase positive staphylococci, particularly Staphylococcus aureus, from food, cosmetic, and clinical samples. Each sterile plate contains 20 mL of nutrient agar enriched with sodium pyruvate and glycine to promote staphylococcal growth. Selective agents such as lithium chloride and potassium tellurite inhibit competing microorganisms while allowing target bacteria to develop. The egg yolk emulsion enables detection of lecithinase activity, producing a clear halo around colonies. Staphylococcus aureus reduces tellurite to form characteristic grey to black colonies surrounded by this halo, creating a distinctive morphology that allows rapid visual identification and differentiation from other staphylococci or background flora during routine microbiological analysis.
