AM-5308 is a KIF18A inhibitor (IC50 47 nM) that also inhibits KIF19A (IC50 0.224 μM) and KIF18B (IC50 17.73 μM) motor activity. It activates mitotic checkpoints, regulating cell division processes such as chromosome segregation and spindle assembly. Demonstrating antitumor activity, AM-5308 shows in vitro cytotoxicity in human bone marrow cells and arrests the cell cycle of MDA-MB-157 cells in mitosis (EC50 0.041 μM). In vivo, it inhibits tumor growth in mouse OVCAR-3 xenograft models.
