MP Biomedicals Betaine (5 M, 2 mL) is a ready-to-use PCR-grade reagent formulated to enhance nucleic acid amplification performance, particularly for GC-rich or structurally challenging templates. As a methylated glycine analogue, betaine modulates DNA duplex stability by minimizing secondary structures, facilitating primer annealing, and improving polymerase processivity under adverse conditions. This additive is particularly useful when standard PCR conditions yield poor specificity or low yields due to problematic regions such as high GC content or stable hairpins. It functions by equalizing the thermal denaturation behavior of AT- and GC-rich sequences and by stabilizing the DNA polymerase in variable local environments of duplex melting. Importantly, the reagent is supplied as a sterile or non-sterile (as indicated in product specs) 5 M aqueous solution that may be added directly to PCR mixes without further dilution, saving preparation time.
