Bile Esculin Agar prepared as a 4 mL slant is a selective and differential medium used for the rapid presumptive identification of Group D Streptococci and Enterococci. Each tube contains nutrient agar enriched with beef extract, peptones, bile salts, esculin, and ferric citrate. The slant configuration provides an expanded inoculation surface while conserving incubator space, making it efficient for biochemical testing and organism maintenance. The medium functions by detecting the ability of bacteria to hydrolyze esculin in the presence of bile. When hydrolysis occurs, esculetin reacts with ferric citrate in the medium to produce a dark brown or black coloration along the slant. This reaction provides a clear visual indicator used in clinical, food safety, and microbiological identification workflows.
