Biotin-16-UTP is a biotin-labeled nucleotide analog, which can be incorporated via a variety of RNA polymerases, including SP6, T3 and T7 RNA polymerases to form labeled RNA. Since T7 RNA polymerase does not significantly discriminate between the natural and the biotinylated nucleotide, the number of biotinylated positions per RNA transcript can be controlled by adjusting the ratio of the biotinylated nucleoside triphosphate (NTP) to the naturally occurring NTP in the transcription reaction. The biotin-labeled RNA can then be detected by a reporter molecule attached to streptavidin, avidin, or anit-biotin antibody. Biotinylated RNA probes are useful as a nonradioactive label in a number of applications including chromosome mapping, in situ hybridization, as well as microarray hybridization.
