Avidin-biotin systems have many applications in immunology and histochemistry. Interactions between avidin and biotin are remarkably strong with dissociation constants on the order of 10-15 M. With an antigen-antibody complex and a biotin-labeled antibody, colorimetric/fluorometric detection of the antigen can be performed using enzymes, fluorophore-labeled avidin or streptavidin. Maleimide biotins react with thiol compounds at a pH of 7-7.5. Maleimide reacts with the sulfhydryl group to create a thioether bond. Unlike other maleimide biotin reagents, Biotin-PE-maleimide can be solubilized in PBS at pH 7.4 to prepare a 2 mM solution without using an organic solvent. The reactivity of maleimide with sulfhydryl groups is higher than that of bromoacetamide, so the required concentration of maleimide biotin is much lower than that of bromoacetamide biotins. Stock solutions of Biotin-PE-maleimide and Biotin-PEAC5-maleimide in DMSO are stable for 1 year at -20°C.
