Lens culinaris agglutinin (LCA) recognizes sequences containing α-linked mannose residues and additional sugars as part of the receptor structure, giving it a narrower specificity than concanavalin A. An α-linked fucose residue attached to the N-acetylchitobiose portion of the core oligosaccharide significantly enhances affinity. This narrower specificity can be exploited for the subfractionation of glycoproteins and glycopeptides with LCA after their initial isolation with concanavalin A. This conjugate features a ratio of biotin to lectin protein that provides optimal avidin/streptavidin and lectin binding activity. Supplied essentially free of unconjugated biotins, it is preserved with sodium azide.
