This kit provides a two-color fluorescence staining of live (green) and dead bacteria (red) using the probes DMAO and Ethidium homodimer III (EthD-III) respectively. DMAO is a green-fluorescent (Ex/Em 496/528 nm) nucleic acid dye staining both live and dead bacteria, whereas EthD-III, a red-fluorescent (Ex/Em with DNA 522/593 nm) nucleic acid dye selectively stains dead bacteria with damaged cell membranes. The assay is simple, rapid, no-wash and economic with dual detection of viability and cytotoxicity simultaneously. The principles are general and applicable to most bacterial types, and analysis is compatible with flow cytometers and fluorescence microscopes using standard settings for fluorescein and propidium iodide. The kit is suitable for fluorescence microscopy (1000 assays) or flow cytometry (100 assays).
