The addition of defibrinated sheep blood supplies essential nutrients and growth factors while allowing visualization of hemolytic reactions produced by different bacteria. These plates enable differentiation of alpha, beta, and gamma hemolysis, which supports preliminary identification of organisms such as Streptococcus and Staphylococcus species. Each sterile 100 x 15mm Petri dish contains approximately 20mL of agar, providing consistent depth and surface moisture for reliable colony development and streaking performance.
