BRD9500 is an orally active phosphodiesterases 3 (PDE3) inhibitor, functioning as a DNMDP analog. It potently and selectively inhibits PDE3A (IC50 = 10 nM) and PDE3B (IC50 = 27 nM), exhibiting antitumor activity by inducing PDE3A/B interactions with SFLN12. It shows an EC50 of 1 nM for SK-MEL-3 melanoma cell line viability and an EC50 of 1.6 nM for HeLa cell viability. In vivo studies demonstrate that BRD9500 inhibits SK-MEL-3 melanoma growth in mice and achieves high plasma levels, making it a valuable candidate for xenograft testing.
