Brilliant Green Agar with Nalidixic Acid is a selective and differential medium used for isolating Salmonella species, excluding Salmonella Typhi, from samples with high background flora. Each sterile 100 × 15 mm plate contains 20 mL of Brilliant Green Agar base supplemented with nalidixic acid to enhance selectivity. The dye brilliant green inhibits gram positive bacteria and many gram negative competitors, while the antibiotic further suppresses organisms such as Proteus and Escherichia coli. Lactose and sucrose fermentation reactions are monitored using phenol red as a pH indicator. Salmonella species generally do not ferment these sugars and form pink to white colonies surrounded by a red medium. This characteristic appearance allows microbiologists to differentiate Salmonella from fermenting organisms during food safety and environmental testing.
