Buffered Charcoal Yeast Extract Agar (BCYE) is the primary enrichment medium used for the isolation and cultivation of Legionella species, including Legionella pneumophila. Each plate contains 20 mL of a yeast extract based nutrient medium buffered with ACES and supplemented with activated charcoal. The formulation includes essential growth factors such as L cysteine and ferrous pyruvate, which are required for Legionella metabolism and replication. Activated charcoal absorbs toxic metabolic by products and peroxides that could otherwise inhibit growth. BCYE agar is widely used for testing respiratory specimens and environmental water samples in clinical and public health laboratories. Colonies typically develop a distinctive ground glass appearance with white to grey coloration, enabling microbiologists to identify and recover Legionella organisms for further confirmation and epidemiological investigation.
