CBL0137 hydrochloride is a potent inhibitor of the histone chaperone FACT, known to activate p53 and inhibit NF-κB. It has demonstrated significant activity in vitro, leading to the absence of living cells at concentrations above 2.5 μM, and enhancing the efficacy of gemcitabine in pancreatic cancer cell lines. In vivo studies show it suppresses tumor growth in various xenograft models, including colon, renal cell carcinoma, and melanoma, both as monotherapy and in combination with gemcitabine.
