CD152, also known as CTLA-4, is a key molecule that functions to inhibit activation of T cells and antigen-presenting cells (APCs). CD152 is a glycoprotein with structural similarities to CD28, in that it shares 20% sequence homology including the ligand-binding motif MYPP. Similar to CD28, CD152 binds CD80 (B7–1) and CD86 (B7–2), but with higher affinity than CD28. CTLA-4 appears to inhibit immune responses by several mechanisms and is a critical mediator of peripheral tolerance. Recent studies have shown that CTLA-4 is expressed by a subset of regulatory T cells (Tregs).
