CD38 inhibitor 1 (compound 78c) is a potent inhibitor of CD38, with IC50 values of 7.3 nM for human CD38 and 1.9 nM for mouse CD38. It also has a Ki of 0.3 nM for wild-type human CD38. In vivo studies showed that a 30 mg/kg oral administration of CD38 inhibitor 1 significantly increased NAD levels in the liver and muscle of diet-induced obese C57Bl6 mice at both 2 and 6-hour time points.
