Cyclin-dependent kinase 1 (CDK1) and its regulatory partner Cyclin B are fundamental to the control of cell cycle transitions, specifically the G2/M phase transition, which ultimately leads to cell division. CDK1, a serine/threonine kinase, forms a complex with Cyclin B necessary for the initiation of mitosis. This complex phosphorylates various substrates to facilitate structural changes required for mitosis. The CDK1/Cyclin B complex is tightly regulated through a series of phosphorylation and dephosphorylation events that control its activation state. The nuclear-cytoplasmic shuttling of Cyclin B1, coupled with CDK1 activity, is crucial for proper mitotic entry and progression. Interestingly, CDK1/Cyclin B1 also plays a role in the coordination of mitochondrial bioenergetics, indicating a broader role in cell metabolism and division. Allosteric modifications, such as acetylation of specific lysine residues, notably lysine-33, also influence CDK1/Cyclin B interaction and function, emphasizing the intricate regulatory mechanisms governing their activity.
