This gene encodes a cyclin-dependent kinase inhibitor that binds to and inhibits cyclin-CDK2 and cyclin-CDK4 complexes, regulating cell cycle progression at G1. Its expression is controlled by the tumor suppressor p53, mediating p53-dependent G1 arrest in response to stress. The protein interacts with proliferating cell nuclear antigen, aiding S phase DNA replication and repair. Cleavage by CASP3-like caspases activates CDK2 and may trigger apoptosis. Mice lacking this gene can regenerate damaged tissue. Multiple alternatively spliced variants of this gene exist.
