The Cereblon (CRBN) complex is an E3 ubiquitin ligase that drives ubiquitination and proteasomal degradation of target proteins. CRBN may also influence memory and learning through interactions with KCTN1. It is widely used in PROTAC drug design and protein degradation studies, where CRBN functions as a substrate adaptor that provides specificity without catalytic activity. CRBN associates with DDB1, Cul4A, and RBX1 to form an active CRBN‑DCX E3 ligase, in which Cullin‑RBX catalyzes ubiquitin transfer from the E2 enzyme to substrates. This complex is validated for enzymatic ubiquitination assays, PROTAC‑mediated ubiquitination studies, and biophysical or HTRF assays analyzing ternary complex formation. Additionally, its FLAG‑tagged components make it suitable for pulldown assays.
