CHS-828 (GMX1778) is a competitive inhibitor of nicotinamide phosphoribosyltransferase (NAMPT) with an IC50 of less than 25 nM. It decreases cellular NAD+ levels, exerting a cytotoxic and potent anticancer effect. The compound increases intracellular reactive oxygen species (ROS) in cancer cells by elevating superoxide levels without inducing ROS in normal cells. Its ROS-inducing effects can be modulated by nicotinic acid in a NAPRT1-dependent manner.
