Type IV collagen is a key structural component of basement membranes consisting of three alpha subunits encoded by six genes (alpha 1-6) which form a triple helix This gene encodes alpha 3 and mutations in it are linked to Goodpasture syndrome where autoantibodies target the collagen in alveoli and glomeruli The epitopes for these autoantibodies are located in the C-terminal domain which is also phosphorylated by a specific kinase during pathogenesis Mutations in this region are associated with autosomal recessive Alport syndrome The gene is arranged in a head-to-head configuration with another type IV collagen gene sharing a common promoter
