DNase Test Agar with Methyl Green is a differential microbiological medium used to detect the production of deoxyribonuclease (DNase) enzymes by bacteria. Each sterile petri dish contains approximately 20mL of nutrient agar incorporating polymerized DNA and the indicator dye methyl green. The dye forms a stable complex with intact DNA, producing a green colored medium. When microorganisms that produce DNase grow on the surface, the enzyme breaks down the DNA molecules surrounding the colony. As the DNA degrades, the methyl green is released, creating a clear halo around DNase positive colonies. This reaction allows rapid visual differentiation of organisms such as pathogenic Staphylococcus species and certain Gram negative bacteria in clinical microbiology laboratories.
