DOPE (Dioleoylphosphatidylethanolamine) is a neutral helper lipid that combines with cationic phospholipids to improve the transfection efficiency of naked siRNA, and it is also an endogenous metabolite. In studies, DOPE-containing liposomes (LNP) exhibited a knock-down (KD) effect on macrophages and dendritic cells and showed fusogenicity. LNP/siRNA significantly suppressed CD45 protein expression, demonstrating potential as an efficient siRNA delivery system for both murine and human macrophages and dendritic cells.
