DTNB (69-78-3) is a biochemical reagent that reacts specifically with free sulfhydryl (-SH) groups It is utilized to quantitatively detect thiol groups in biological samples DTNB exerts its activity by undergoing disulfide bond cleavage upon interaction with free thiols leading to the formation of a colored derivative 5-thio-2-nitrobenzoate (TNB) which is measurable by spectrophotometry at 412 nm Based on these properties DTNB is widely applied in the quantification of reduced glutathione (GSH) and assessment of thiol levels in tissues blood and cultured cells enabling studies of oxidative stress redox state and thiol-reactive drug screening
