The DTNB (Ellman's) Thiol Oxidative Stress Assay Kit is designed to measure free thiol groups that arise due to oxidative stress, which is an underlying component of acute and chronic kidney disease. Exocells kit uses Ellman's Reagent otherwise known as DTNB or 5-5'-dithiobis[2-nitrobenzoic acid] which reacts with the disulfide bond in a sample to produce 2-nitro-5-thiobenzoic acid. Under conditions of oxidative stress, free thiols decrease and disulfides increase. Determination of the free thiol concentration in biological samples reflects the ability to detoxify lipid (and other) peroxides. To complete the assay, aliquots of standard or sample are reacted with rehydrated Ellman’s reagent (DTNB) in microtiter wells. Absorbance is read at 405 nm using a reference filter of 630 nm. Concentration in samples is determined from the standard curve.
