EC Broth with Durham Tube is a selective enrichment medium used for detecting fecal coliforms and Escherichia coli in water, food, and environmental samples. Each tube contains approximately 10mL of buffered lactose broth supplemented with bile salts that inhibit Gram positive bacteria and many non coliform organisms. An inverted Durham tube is included to capture gas produced during lactose fermentation. When inoculated samples are incubated at elevated temperatures around 44.5°C, fecal coliforms ferment lactose and release gas that becomes trapped as a visible bubble in the Durham tube. The broth typically becomes turbid as bacterial growth occurs. This elevated temperature test helps distinguish fecal coliforms from other environmental coliform bacteria.
