EC Broth with MUG and Durham Tube is a selective detection medium designed for confirming the presence of Escherichia coli in environmental, food, and water samples. Each tube contains 10mL of EC broth supplemented with the fluorogenic substrate MUG (4 methylumbelliferyl β D glucuronide) and an inverted Durham tube for gas detection. During incubation at approximately 44.5°C, fecal coliform organisms ferment lactose and produce gas that accumulates in the Durham tube. If E. coli is present, the enzyme β glucuronidase cleaves the MUG substrate, releasing a fluorescent compound. When exposed to long wave ultraviolet light, positive tubes exhibit a bright blue fluorescence. This dual detection system allows rapid and reliable confirmation of E. coli in microbiological testing.
