Isolate a carbohydrate-binding lectin protein from an extract of jack bean meal.

Affinity chromatography is applied to the purification of biologically active molecules. A ligand is coupled to an insoluble support. The specific protein binds to the ligand, which results in the separation of a specific protein from a mixture.

• Students prepare a seed extract, fractionate the extract by affinity chromatography and elute the bound glucose-binding protein
• As a second experiment, the presence of biological activity is determined by an immunoblot enzyme assay
• Requires two hours
