Product DescriptionELISA reaction buffer has a 50 mM concentration of carbonate-bicarbonate and a pH level of 9.6. It contains 1.59 g Na2CO3, 2.93 g NaHCO3, and dissolved in 1 liter of deionized water.Carbonate/bicarbonate buffer at a pH greater than 9 is used for the attachment of an antigen or of an antibody onto an ELISA plate. Direct coating of ELISA is an antigen immobilization technique, in which antigens are directly attached to the plate by passive adsorption. Most but not all proteins bind tightly to the polystyrene surface of microplates in alkaline conditions. Indirect coating of ELISA is another antigen immobilization technique in which antigen-specific antibodies are adsorbed onto the plastic, which in turn binds and immobilizes the antigen upon incubation with the antigen sample. Both techniques typically use the same buffer.
