Embelin is a novel cell-permeable inhibitor of XIAP (X-linked inhibitor of apoptosis) which was discovered by screening a library of natural products derived from traditional Chinese medicine. It binds to the XIAP BIR3 domain preventing its binding to and inhibition of caspase-9 and effector caspases. Embelin selectively inhibits cell growth in cancer cells expressing high levels of XIAP (IC50 = 3-6 µM) versus normal cells with low levels of XIAP. Alternative name: Embelic acid.
