Matrix metalloproteinases (MMPs) are a family of enzymes that are responsible for the degradation of extracellular matrix components such as collagen, laminin and proteoglycans. All MMPs share: sequence homology, a catalytic mechanism dependent upon a zinc ion at the active center, ability to cleave one or more extracellular matrix components, secretion as zymogens which are activated by removal of an approximately 10kDa segment from the N-terminus, and regulation by endogenous inhibitors. These enzymes are involved in normal physiological processes as well as in metastasis. MMP-2 and MMP-9 are members of the gelatinase family and elevated levels may be correlated with metastatic potential. MMP-1 is also known as interstitial collagenase and MMP-3 as stromelysin-1. Activation and activity of the MMP's are regulated by the TIMP (tissue inhibitors of metalloproteinase) family members. TIMP-1 preferentially regulates MMP-9 and TIMP-2 regulates MMP-2.
