Fluid Thioglycollate Medium (FTM) is a standard liquid culture medium used for sterility testing in pharmaceutical and clinical laboratories. Each tube contains 21.5mL of broth prepared in accordance with United States Pharmacopeia (USP <71>) specifications. The medium supports the growth of a wide range of microorganisms, including obligate aerobes, microaerophiles, and strict anaerobes. Sodium thioglycollate and L cystine function as reducing agents that lower the oxidation reduction potential, allowing anaerobic organisms to grow in the deeper portions of the tube while aerobic organisms grow near the surface. A small amount of agar stabilizes the oxygen gradient by preventing convection currents. Resazurin is included as an oxidation indicator, producing a pink coloration in oxygen rich areas while deeper layers remain clear.
