Concanavalin A (ConA) recognizes α-linked mannose residues present as part of a "core oligosaccharide" in many serum and membrane-bound glycoproteins. At neutral and alkaline pH, Con A exists as a tetramer, but below pH 5.6, it dissociates into dimers of 52 kDa. Derivatizations can produce stable forms with dimeric structures. Con A requires calcium or manganese ions at each binding site, so avoid using buffers that bind calcium (such as phosphate) to dilute Con A since a gradual loss in activity may occur. This fluorescein-labeled Con A features a ratio of fluorophores to lectin protein that provides optimal staining (excitation 495 nm, emission 515 nm). Supplied as a solution essentially free of unconjugated fluorophores, it is preserved with sodium azide. The recommended inhibiting/eluting sugar is a mixture of 200 mM α-methylmannoside/200 mM α-methylglucoside.
