Griffonia simplicifolia lectin II (GSL II) is a dimeric glycoprotein composed of two subunits of nearly identical size. Each subunit has disulfide-linked chains and a binding site for α- or β-linkedN-acetylglucosamine residues. Unlike other N-acetylglucosamine-specific lectins, increasing the number of N-acetylglucosamine residues beyond two does not improve affinity. GSL II is unique in its ability to recognize exclusively α- or β-linked N-acetylglucosamine residues on the nonreducing terminal of oligosaccharides. This fluorescein-labeled GSL II features a ratio of fluorophores to lectin protein that provides optimal staining (excitation 495 nm, emission 515 nm). Supplied as a solution essentially free of unconjugated fluorophores, it is preserved with sodium azide. The recommended inhibiting/eluting sugar is chitin hydrolysate or 200 mM N-acetylglucosamine.
