Forskolin is a cell-permeable labdane diterpene derived from Coleus Forskohlii which directly stimulates adenylate cyclase (AC). This unique diterpene increases in levels of intracellular cyclic AMP (cAMP) from ATP in various eukaryotic cells by activating AC. cAMP is a second messenger mediating a variety of signaling pathways and is related to lipolysis via thermogenesis. It is reported that forskolin induces CYP3A gene expression in primary hepatocytes by functioning as agonists of the pregnane X receptor (PXR). Protein kinase A (PKA) signal transduction pathway activated by cAMP enhances CYP3A gene expression induced by PXR.This compound is a potent inhibitor of platelet aggregation and proven for antitumor effects. cAMP signaling related to forskolin is relevant to the effects in induction of mesenchymal-to-epithelial transition, inhibition of cell proliferation and metastasis. In addition, it helps sensitivity to antitumor drugs enhanced in cancer cells.
