Description
The Ion Torrent Genexus Dx Purification System is an in vitro diagnostic (IVD) platform that delivers a simplified user experience for clinical labs. The system automates nucleic acid extraction, purification, and quantification from clinical specimens for use in next-generation sequencing (NGS) applications in as little as two hours. Consumables for the system come prefilled with reagents and can support up to 12 samples per run. The system includes an onboard Qubit fluorometer to automate post-extraction nucleic acid quantification.

The Genexus Dx Purification System automates the extraction and quantification of nucleic acids from FFPE samples for use on the Genexus Dx Integrated Sequencer. The Genexus Dx Software supports running IVD protocols and assay development protocols for clinical research. With a single touch point, the system requires just 10 minutes of hands-on time. Using the Genexus Dx FFPE DNA/RNA Purification Kit, the purification system can extract both DNA and RNA sequentially from a single formalin-fixed, paraffin-embedded (FFPE) lysate.
Prefilled reagents
The Genexus Dx Purification System consumables are prefilled with most reagents necessary to perform the extraction, so only a few pipetting steps are required per sample to set up the run. Simply remove the foil seal, add enzymes and your samples, then place the consumables onto the system.
Vision system
Just like the Genexus Dx Integrated Sequencer, the Genexus Dx Purification System uses an onboard vision system to verify reagent placement and detect errors in real time. The vision system continuously verifies the consumables to help ensure they are appropriate to the purification run type and helps prevent the use of expired or previously used consumables. Feedback on the consumables loaded in the system is displayed on the system's touchscreen.
MagMAX technology
MagMAX technology uses magnetic beads to perform nucleic acid extraction. MagMAX beads offer superior binding capacity due to their large surface area, accept a greater range of samples with differing viscosities, and allow for efficient elution in small volumes.
Automated quantification
Quantification of extracted nucleic acids is performed by an onboard Qubit fluorometer. Two dyes are used to measure the concentration of DNA or RNA from each extracted sample. Automating post-run quantification saves hands-on time and allows for immediate use of the extracted samples in genetic analysis workflows after run completion.
Review concentrations
Nucleic acid concentrations can be reviewed directly on the system's touchscreen. Alternatively, the data can also be exported to a USB drive for further review and downstream run planning.
48‐well nucleic acid archive plate
After extraction, the quantified nucleic acids are transferred directly to the 48‐well archive plate and kept cool for up to one hour after run completion. The 48‐well nucleic acid archive plate is barcoded to facilitate the tracking of sample extractions across several stored archive plates.
Rapid turnaround
Run times on the Genexus Dx Purification System range from 2 to 5.5 hours depending on the number of samples extracted and the selected protocol. Automated quantification helps significantly reduce post-run hands-on time, allowing for rapid transition to subsequent genetic analysis workflows.
Post-run clean
At the completion of each run, all used consumables are discarded by the operator, which is followed by an automated UV cleaning step using UV light to decontaminate the deck. This automated UV cleaning minimizes potential run-to-run carryover of nucleic acid, which could lead to contamination of downstream workflows.
Intended use
The Genexus Dx Purification System is a sample preparation instrument that automates nucleic acid extraction, purification, and quantification of DNA and RNA from FFPE tissue sections for use in NGS applications. The system uses MagMAX magnetic bead-based technology to perform nucleic acid extraction. Purified nucleic acid is quantified using the onboard Qubit fluorometer, which uses fluorescence to measure nucleic acid.
