GNE-317 is a PI3K/mTOR inhibitor that can cross the blood-brain barrier (BBB). It is an oxetane derivative of GDC-0980, synthesized to reduce substrate affinity for efflux transporters. While GNE-317 shows greater delivery and enhanced therapeutic targeting efficacy compared to GDC-0980, it showed limited efficacy in inducing cell death in GL261 cell lines in vitro and was not found effective in vivo in GL261 tumor models.
