GO-203 TFA is a potent inhibitor of the MUC1-C oncoprotein. It is an all D-amino acid peptide that contains a poly-R transduction domain linked to a CQCRRKN motif, which binds to the MUC1-C cytoplasmic tail and prevents MUC1-C homodimerization. This compound downregulates TIGAR protein synthesis by inhibiting the PI3K-AKT-S6K1 pathway. It also induces the production of Reactive Oxygen Species (ROS) and leads to a loss of mitochondrial transmembrane potential. GO-203 TFA has been shown to inhibit the growth of colon cancer cells both in vitro and as xenografts in nude mice.
