Gram Negative (GN) Broth is a selective enrichment medium designed for the recovery and cultivation of Gram negative enteric pathogens, particularly Salmonella and Shigella species. Each tube contains 10mL of broth prepared using Hajna’s modification, which enhances the growth of target organisms while suppressing competing bacteria. The medium achieves selectivity through inhibitory agents such as sodium citrate and sodium deoxycholate that reduce the growth of Gram positive organisms and many coliform bacteria. Mannitol serves as a preferred carbohydrate source that supports the rapid multiplication of Salmonella and Shigella during the enrichment phase. This short enrichment step increases pathogen numbers before plating onto selective agar media for isolation. GN Broth is widely used in clinical, food safety, and environmental microbiology laboratories.
