Cayman’s HDAC8 Inhibitor Screening Assay provides a convenient fluorometric method for screening HDAC8 inhibitors in a two-step reaction. In the first step, the substrate, which is comprised of the p53 sequence Arg-His-Lys-Lys(ε-acetyl)-AMC, is incubated with recombinant human HDAC8. Deacetylation sensitizes the substrate such that treatment with the developer in the second step releases a fluorescent product. The fluorophore can be EAsily analyzed using an excitation wavelength of 360 nm and an emission wavelength of 460 nm.
