Horse Blood Overlay Agar (Bilayer) is an enriched culture medium designed to improve visualization of hemolytic reactions and support the growth of fastidious microorganisms. Each plate features a two layer design consisting of a nutrient rich base agar layer and a thin overlay containing 5–7% defibrinated horse blood. This bilayer structure enhances contrast, allowing hemolysis zones produced by bacterial colonies to be observed more clearly than on standard blood agar plates. Horse blood is often preferred for certain diagnostic applications because it lacks the enzyme NADase, preserving essential growth factors such as Factor V required by organisms like Haemophilus species. The medium supports isolation and differentiation of hemolytic bacteria by producing clear alpha or beta hemolytic reactions, making it useful in clinical microbiology and diagnostic laboratories.
