Frequently Asked Questions (FAQs)
If you use Alexa Fluor 488/Alexa Fluor 555 dye-conjugated secondary antibodies and you have a Cy5/Alexa Fluor 647 filter set, you may use a far-red nucleic acid stain, such as NucRed Dead 647 ReadyProbes Reagent (Cat. No. R37113) or SYTOX Deep Red Nucleic Acid Stain (Cat. No. S11380). Do not use these far-red nucleic acid stains if using an Alexa Fluor 594 dye-conjugated secondary antibody as there is too much spectral overlap of Alexa Fluor 594 into the Cy5/Alexa Fluor 647 range.
We do not recommend using the Human Cardiomyocyte Immunocytochemistry Kit for flow cytometry analysis. The kit and protocol were specifically developed for imaging analysis (microscopy/high-content screening (HCS)).
Only the NucBlue Fixed Cell ReadyProbes Reagent may be purchased separately as Cat. No. R37606. We do offer the secondary antibodies separately, but not in the same proprietary concentration as provided in the kit. We also offer formaldehyde, DPBS, BSA, and saponin as separate catalog numbers, but not in the volumes or as formulated in the kit. The primary antibodies are not available as separate stock items; all non-stock items in the kit may be available as a custom order. Please contact your Account Representative.
Cardiomyocytes generated using PSC Cardiomyocyte Differentiation Kit have been tested for key markers such as TNNT2, Nkx2.5, MYH6, and Alpha-Actinin. The Cardiomyocyte Immunocytochemistry Kit (Cat. No. A25973) contains validated antibodies to measure TNNT2 and Nkx2.5 in cultures generated using PSC Cardiomyocyte Differentiation Kit.
