The Human Cardiomyocyte Immunocytochemistry Kit enables optimal image-based analysis of two key cardiomyocyte markers: NKX2.5 and TNNT2/cTnT. It is the only kit that offers superior imaging for cardiomyocytes in one box, with a complete set of primary and secondary antibodies, a nuclear DNA stain, and all of the pre-made buffers for an optimized staining experiment.

    The antibodies included in the kit have been carefully selected and validated for high performance in the immunocytochemistry analysis of cardiomyocytes. The antibodies are well suited for the characterization of cardiomyocytes generated from pluripotent stem cells using the Gibco™ PSC Cardiomyocyte Differentiation Kit.

    The Human Cardiomyocyte Immunocytochemistry Kit provides the NucBlue Fixed Cell Stain. If I don't have the correct filter set for this stain, can I use another nucleic acid stain?

    If you use Alexa Fluor 488/Alexa Fluor 555 dye-conjugated secondary antibodies and you have a Cy5/Alexa Fluor 647 filter set, you may use a far-red nucleic acid stain, such as NucRed Dead 647 ReadyProbes Reagent (Cat. No. R37113) or SYTOX Deep Red Nucleic Acid Stain (Cat. No. S11380). Do not use these far-red nucleic acid stains if using an Alexa Fluor 594 dye-conjugated secondary antibody as there is too much spectral overlap of Alexa Fluor 594 into the Cy5/Alexa Fluor 647 range.

    Can I use the Human Cardiomyocyte Immunocytochemistry Kit for flow cytometry analysis?

    We do not recommend using the Human Cardiomyocyte Immunocytochemistry Kit for flow cytometry analysis. The kit and protocol were specifically developed for imaging analysis (microscopy/high-content screening (HCS)).

    Do you offer the individual components of the Human Cardiomyocyte Immunocytochemistry Kit as standalone products?

    Only the NucBlue Fixed Cell ReadyProbes Reagent may be purchased separately as Cat. No. R37606. We do offer the secondary antibodies separately, but not in the same proprietary concentration as provided in the kit. We also offer formaldehyde, DPBS, BSA, and saponin as separate catalog numbers, but not in the volumes or as formulated in the kit. The primary antibodies are not available as separate stock items; all non-stock items in the kit may be available as a custom order. Please contact your Account Representative.

    I am planning to use the PSC Cardiomyocyte Differentiation Kit. Which markers can I expected my differentiated cells to express?

    Cardiomyocytes generated using PSC Cardiomyocyte Differentiation Kit have been tested for key markers such as TNNT2, Nkx2.5, MYH6, and Alpha-Actinin. The Cardiomyocyte Immunocytochemistry Kit (Cat. No. A25973) contains validated antibodies to measure TNNT2 and Nkx2.5 in cultures generated using PSC Cardiomyocyte Differentiation Kit.
