Interlukin-1 polypeptides are pleiotropic cytokines exerting diverse functions in different tissues. IL-1R1 (also known as CD121a) is a receptor for interleukin-1 alpha (IL-1α), interleukin-1 beta (IL-1β), and interleukin-1 receptor antagonist (IL-1Ra). Human IL-1R1 was first cloned from human T cells. This receptor contains a large cytoplasmic region and an extracellular, IL-1 binding portion composed of three immunoglobulin-like domains. IL-1R1 expression has been found in many cell types including T cells, macrophages, neutrophils, eosinophils, basophils, mast cells, fibroblasts, and endothelial cells. IL-1R1 can be shed from the cell membrane as a soluble form (sIL-1R1) which can function as a soluble decoy receptor and prevent the binding of IL-1α, IL-1β, and IL-1Ra to membrane IL-1R1.
