Use brain-derived neurotrophic factor (BDNF) to activate AKT and ERK pathways (Mattson et al. Ageing Res Rev, 2004). In combination with glial cell line-derived neurotrophic factor and other supplements, BDNF can be used to differentiate human pluripotent stem cell (hPSC)-derived neural progenitor cells into neurons (Brafman. Methods Mol Biol, 2015). BDNF also acts as a survival factor for human embryonic stem cells when plated on feeder cells or Corning Matrigel (Pyle et al. Nat Biotechnol, 2006). BDNF regulates synaptic transmission and plasticity at adult synapses in the central nervous system, contributing to adaptive neuronal responses (Reichardt. Philos Trans R Soc Lond B Biol Sci, 2006). Human recombinant BDNF from STEMCELL comes lyophilized with ≥ 95% purity; specific activity EC50 ≤ 40 ng/mL (for Lot #1000038506 or higher) and EC50 ≤ 2 μg/mL (for other lots); endotoxin levels verified ≤ 1.0 EU/μg protein.
