Use ciliary neurotrophic factor (CNTF) to differentiate human pluripotent stem cell (hPSC)-derived neural progenitor cells into astrocytes (Krencik & Zhang. Nat Protoc, 2011). CNTF binds to its receptor CNFTRα and induces a heterodimer formation with the IL-6 receptor gp130 and LIF receptor (LIFR)-β, and activates the JAK/STAT, ERK, and the PI3K signaling cascades (Schuster et al. J Biol Chem, 2003). CNTF plays an important role in neurogenesis and the differentiation of neural stem cells and has been suggested to possess a therapeutic role in treating neurological disorders (Ding et al. J Alzheimers Dis, 2013.) CNTF also has been reported to protect oligodendrocytes from death induced by apoptosis (Louis et al.). Human recombinant CNTF from STEMCELL is animal component free and comes lyophilized with ≥ 95% purity; specific activity EC50 ≤ 200 ng/mL; endotoxin levels verified ≤ 0.2 EU/μg protein.
