α-Hydroxyglutaric acid disodium is an α-hydroxy acid form of glutaric acid. It is a competitive inhibitor of multiple α-ketoglutarate-dependent dioxygenases, including histone demethylases and the TET family of 5-methylcytosine (5mC) hydroxylases. Research indicates that isocitrate dehydrogenase 1 (IDH1) and IDH2 mutations, common in gliomas and acute myeloid leukemia, lead to altered production of α-ketoglutarate and α-Hydroxyglutaric acid disodium. Furthermore, it has been shown to inhibit the activity of TET 5-methylcytosine hydroxylases and, in U-87MG cells, increases HIF-1α while decreasing endostatin.
