Indole Broth, commonly referred to as Tryptone Water, is a biochemical test medium used to determine the ability of microorganisms to produce indole from the amino acid tryptophan. Each tube contains 4mL of nutrient broth rich in tryptophan containing peptones that serve as substrates for bacterial metabolism. Organisms possessing the enzyme tryptophanase break down tryptophan to produce indole, pyruvic acid, and ammonia during incubation. After incubation, the presence of indole is detected by adding Kovac’s or Ehrlich’s reagent, which reacts with indole to produce a characteristic red layer at the liquid surface. A positive reaction indicates organisms such as Escherichia coli or Proteus vulgaris, while negative reactions remain yellow or unchanged. This test is an essential component of the IMViC series used to differentiate Enterobacteriaceae.
