Determine if Your Instrument is Set Up to Read Tb TR-FRET Assays
Setting up your instrument to read Tb TR-FRET is the first step in running a successful Tb TR-FRET assay. This kit allows you to quickly determine if your instrument is set up properly to read Tb TR-FRET assays. Simply add a small volume into separate wells, place the plate into the instrument and read. If the ratio of high to low control signal is below 2 the instrument may not be setup properly. If the ratio of high to low control signal is between 2 and 4 you can proceed with a Tb TR-FRET.
Use the Kit as a Positive Control in Tb TR-FRET Assays
When running Tb TR-FRET assays, add a small volume of the high and low controls to separate wells, on your assay plate. Place the plate into the instrument and read, according to instructions. A high to low control signal ratio of between 2 and 4 offers you the confidence that the instrument is performing well.

The single most common reason that a TR-FRET assay fails is that an incorrect choice of emission filters was made. Unlike other fluorescent assays, the filters used in a TR-FRET assay must be exactly those that are recommended for your instrument as the emission filter choice can make or break the assay. The excitation filter has more of an impact on the assay window. Please refer to our instrument setup guides in our instrument compatibility portal (https://www.thermofisher.com/instrumentsetup). Note: Test your microplate reader's TR-FRET setup before you begin any work with your assay.
