Use interleukin 17A (IL-17A) to modulate the proliferation of marrow-derived MSCs and suppress osteogenic differentiation and bone formation of bone marrow-derived MSCs (Mojsilović et al. Mediators Inflamm, 2015). IL-17A is a potent pro-inflammatory cytokine that plays a key role in defense against pathogens and activates the NF-κB, MAPK, and C/EBP signal transduction pathways (Gaffen. Nat Rev Immunol, 2009). In healthy mouse bone marrow, IL-17A stimulates myeloid and early stage erythroid progenitor cells but inhibits late stage erythroid progenitor cells (Mojsilovic et al.). IL-17A is produced by T helper 17 cells, CD8+ T cells, γδ T cells, natural killer T cells, B cells, neutrophils, innate lymphoid cells, and mesenchymal stromal cells (Mojsilovic et al.). Mouse recombinant IL-17A from STEMCELL comes lyophilized with ≥ 95% purity; EC50 ≤ 5 ng/mL; endotoxin levels verified ≤ 1 EU/μg protein.
