The SUMO Protease box contains SUMO Protease and buffers. The TOP10 box contains 21 x 50μL aliquots of chemically competent E. coli, S.O.C. medium, and a control plasmid. The One Shot BL21(DE3) box contains 21 x 50μL aliquots of chemically competent E. coli, S.O.C. medium, and a control plasmid.

Yield high levels of protein expression in E. coli

• Uses small ubiquitin-related modifier (SUMO) fusion, belonging to growing family of ubiquitin-related proteins, to enhance solubility of expressed fusion proteins
• Contrast to ubiquitin, SUMO is involved in stabilization and localization of proteins in vivo
• After expression, 11 kd SUMO moiety can be cleaved by highly specific and active SUMO (ULP-1) protease at carboxyl terminal, producing native protein
• Enhanced solubility with N-terminal SUMO fusion
• Highly efficient cleavage produces native protein of interest with SUMO (ULP-1) protease
• Highly specific cleavage eliminates chance of protein of interest being internally digested, regardless of its amino acid sequence
